Eek! Hopefully there is no busted piping. It is unfortunately common for water to leak from upstairs bathroom. There could be various reason. Such as the grout has come away due to movement or/and age. Or water proofing was not done correctly.
Don't forget to check your floor joists for any water damage when you open up the ceiling. Also any load bearing walls if any in the area.
You've got a lot going on which is making you feel unwell. Also the mould is probably making you feel unwell too. I hope everything will get sorted swiftly so you can focus on both you and your wife's health.

As for the plumbing, unfortunately it is some busted pipe but looking at the good side of it, it has busted only after the tap so as long as we don't turn the tap on there is no leak. I've made a new post with photos, we will have to cut the wall behind the two showers to access the pipes for repair.

Just through my work (drafting) and my partner's line of work. My sister had a similar problem as yours. Both showers upstairs were leaking water through the ceiling. My partner had gone to check it out and fixed it up.
