It will be surprise trip for my wife's birthday to the carnival in Venice.

Carnival would definitely be the best time to visit Venice..
Venice is a very small city so you will be able to cover everything on foot in just 2 days. Best way to see the city is to roam without any target in mind.. take any road, any street and you will surprise yourself
The must visit are St. Marks Square , Gondola ride in the Grand Canal and Rialto Bridge.. also I would advise you to get a boat pass, that way you will be able to ride the canal whenever you want!
