Analysis of carbohydrate content in a wheat sample

So in the lab today I was working with some food samples which was given to me by my lecturer,he asked me to find the % of carbohydrates in a wheat sample, so I will be giving a step by step description of what I did, hope it interests you
Carbohydrate content fall under proximate analysis. Proximate analysis is the partitioning of compounds in a food sample into six categories which are carbohydrates, crude fiber, crude protein, lipids, ash and moisture. But today I only worked on carbohydrate. The procedure I used are as follows.
I took 1g of the wheat sample and grind it into fine particles.
I then added the fine particle into a boiling tube and added 10ml of 2.5% sulphuric acid and allowed it to boil at 100oc for 15mins.
After 15mins I filtered the mixture thoroughly with a cotton wool and I made the filtrate up to 250ml with distilled water
I then took 1ml from the mixture into another test tube and added 9ml of distilled water and shake vigorously and then 4ml of authrone reagent was added.
The absorbance was read at 540nm and it's standard is 0.589
Then for the calculation I used absorbance of sample divided by absorbance of standard (0.589) multiplied by 25 divided by weight of sample and thats how you get the % carbohydrate in a food sample. Hope you learnt from this .
