RE: My shittiest method (#002): Examination of free radicals in plant plasma membranes

loool, holy crap.

signal to noise ratio was 2:1 or 3:1 in best case

In other words, your best case was scraping at the LOD. Did your boss dream of quantification too?

Sort:

Quantification of the effect (there is no effect)
Reproducibility of results (with the membranes that were never the same)
Concentration dependency (of the thing without any effect)
And the effect of NAD/NADH & NADP/NADPH (almost no effect)
Additional normalization to protein concentration in membranes

And the same thing with the cell wall isolates that were even worse :D

I mean, just look at the paper grade Figure:

And here you have 2 species, *OH and *OOH (white and black dot). 30 years of financing one big nothing

All the results were: you put the pure O2 atmosphere, *OOH signal rises (wooow...)
HCN kills all the signals (Wooow, seriously?)
and if you put N2, all the signals die (again, sarcastic, wooow...)

Some poor guy came after me, to do the same crapy thesis and he was the fifth (5th) person who couldn't do it. Finally, old boss became retired.

I am so fucking glad that I graduated 500 km farther north ;-)
