Good day ladies and gentlemen. It feels good having you around on this edition of my diary.

MICROBIAL LOAD
It was a wonderful Monday morning. Still very tired, an aftermath of a hectic yesterday.
The sun rose very early today, it was already 7:30am, didnāt feel like waking up, but the sun ray from the window became a motivation.
Said morning prayer as usual, little push up, mouth was taken care of, took a shower and had hot beverage with bread for breakfast.
I have an appointment to catch up by exactly 10am at the College of Natural Science (COLNAS) Michael Okpara University of Agriculture Umudike (MOUAU). Therefore, I took off from my house at exactly 9:30am.
I arrived just in time. Then we started organizing the materials we needed for our practical.
We have done an aspect of this practical some days ago which was Medium and sample preparation
So this time around, we will be doing the concluding aspect which is gram staining or ** smearing**
MATERIALS
ā¢ Wire link
ā¢ Cunicle flask
ā¢ Distilled Water
ā¢ Microscope
ā¢ Slide
ā¢ Fire
ā¢ Inoculated fish sample in a Petri dish
ā¢ Laboratory coat
ā¢ Masking tape
ā¢ Writing materials etc
REAGENTS
ā¢ Primary stain
ā¢ Mordant iodine
ā¢ Colorant
ā¢ Counter stain

10:30am
We kicked off with the procedureā¦..
The reagents were arranged serially in the order I listed them above with distilled water in-between, exampleā¦
ā©Primary stain, ā©water, ā©mordant iodine, ā©water, ā©colorant, and ā©counter stainā¦..
ā”ļø Put your wire link in a fire and wait for it to turn red. Remove it and put it in water, then use it to grab sample from the colonies in the inoculated sample. Return the wire back into the water.

ā”ļø smear the grabbed sample on a slide.
ā”ļø Heat fix the smeared sample on the fire: this is done by passing the slide above a flame until the smeared sample is dried.
ā”ļø Now start dipping the heat sample into the reagents in the order they were arranged, starting from primary stain to the last which is the counter stain.
Dip in each of these reagents for one minute each.

ā¹ļø At the last one which is the counter stain, remove your slide and keep it in the dried chamber for 24hours (1day) before you view with the microscope.
This was how my day went. Thanks for hanging around and I beseech to make it a date with me as we meet in the next 24hours to view our sample under the microscope.
