Cleopatra berhasil merayu Caesar untuk bersama-sama menggulingkan tahta. Ptolemy XII tewas ditenggelamkan di sungai Nil dan Cleopatra naik tahta. Dari percintaan dengan Julius Caesar, ia dikaruniai seorang putra, Ptolemy Caesar, atau Caesarion.

Akhir Tragis Cleopatra

Dalam situasi perang yang berkecamuk, akhrnya kekuasaan Romawi berhasil dikonsolidasi oleh Octavian dan Agrippa, dan Dewan Senat Romawi menngumumkan perang terhadap Mesir, dan meminta Mark Anthony pulang untuk bertugas kembali ke Romawi. Namun karena cintanya yang terlalu dalam terhadap Cleo, Anthony menolak panggilan tugas dan tetap berkhianat terhadap Romawi. Ia dan pasukannya tetap berpihak pada Mesir.

Suasana makin tidak kondusif, dan Anthony memilih jalan mati dengan cara bunuh diri dengan pedangnya. Situasi itu membuat Cleopatra menyimpan dendam dan lupa mendalam. Tetapi situasi politik memang sedang tak memihak kepadanya. Ia akhinya mengambil jalan singkat, dengan mengurung diri di tempat pengasingan, ia bunuh diri dengan membiarkan ular berbisa mematuk tubuhnya. Ia tutup usia menghembus nafas terakhir dalam usia 39 tahun.

Meninggalnya sang dewi Cleopatra, seiring pula dengan berakhirnya kejayaan kerajaan Mesir Kuno. Mesir kemudian takluk di bawah Romawi dan dijadikan salah satu Provinsi Kekaisaran Roma. Cleoptara dan Marks Anthony dikuburkan berdampingan di sebuah piramid.
