RE: How to live healthy? Part 1: What are Caloric Restriction Mimetics (CRM)?

jojo effect

do you mean yo-yo effect?

Rapamycin is therefore very specific and extremely effective, even nanomolar doses show clear effects

This is some nice data, do you have a dose response showing at what concentration of rapamycin the onset of phosphorylation inhibition occurs in cells? I am curious what the IC50 is.

raises the question whether a radical low-carb diet and other strategies might eventually be very unhealthy.

Perhaps it is really the extreme high-fat of the atkins diet that eventually catches up with you. Maybe fat shuttling processes get overwhelmed? Do people on atkins diets typically have super high VLDL levels? Have there been any studies that track VLDL over time for people on these diets? Maybe its the triglycerides that get ya?

optimal dosage is also a problem

What is the optimal dose of 2-DG to achieve a response? If its too high then its likely truly not clinically relevant. Nobody wants to eat a burrito of something for a treatment. :D

n worms it was shown that 2-DG causes oxidative stress and a longer lifespan. However, the addition of vitamin C and E prevents these effects immediately. Therefore, keep your fingers off vitamin supplementation

Dunno if I would go that far. Keep your fingers off vitamin supplementation unless you are low on those respective vitamins. You aren't going to live too long with scurvy!

So if you ask me if metformin is good for you or not, all I just can say is that sports are really helpful.

Its probably not! Certainly the drawbacks are outweighed for people with diabetes though. And yeah... exercise definitely wins over a pharmacological intervention when ever possible :p

In fact, an increases in lifespan was observed in mice after administration of acarbose [23]. Unfortunately, studies show that this is primarily the case for males [25].

Look for information on acarbose pharmacodynamics, perhaps the difference in liver cips between male and female mice explain that data.

trans-reseveratrol

Nah, its got really really bad bioavailability! There are some studies which indicate that its metabolites may also have an effect, but I wouldn't really put a lot of stock into it really having strong in vivo efficacy for much of anything.

You did a lot of nice research here, really well done job!
